My Mutant Rat Dogs

Back in July of '04, I mentioned that I'm probably the "first professionally trained commercial artist to have a peer reviewed paper on opthalmology on her resume".

The research has been continuing, and the team has just published another, this time in Investigative Ophthalmology and Visual Science. If you have any background in molecular genetics, you may find the abstract rather interesting.;

White blood cells of affected dogs contained less than 30% of the normal amount of two specific mtDNA sequences, compared with the content of the nuclear-encoded glyceraldehyde-3-phosphate dehydrogenase (GA-3-PDH) reference gene. Retina and RPE tissue from affected dogs had reduced mRNA transcript levels for the two mitochondrial genes detected in the RDA experiment. Transcript levels for another mtDNA-encoded gene as well as the nuclear-encoded mitochondrial Tfam transcription factor were reduced in these tissues in affected dogs. Mitochondria from affected dogs were reduced in number and size and were unusually electron dense.

What's interesting is that the retinal dysplasia affected dogs are lacking normal mitochondria DNA in all the cell types tested so far - and that they are not only alive, but otherwise healthy and fertile.

THREE animal rights activists were jailed for 12 years each and a fourth imprisoned for four years for a terror campaign against an English guinea pig farm that culminated in a grave robbing.

John Ablewhite, Kerry Whitburn and John Smith were today each handed 12-year prison terms at Nottingham Crown Court in central England after pleading guilty to a charge of conspiring to blackmail the farm owners.

Josephine Mayo was jailed for four years after admitting a lesser part in the six-year campaign against the Hall family who own a farm in Staffordshire, central England, which bred the rodents for medical research purposes.

What began as a peaceful protest outside the gates of Darley Oaks Farm in the hamlet of Newchurch in 1999 escalated into threats, intimidation and even violence against the owners and any person or business linked to it.

That included a pedophile smear campaign, death threats, hate mail, malicious phone calls, hoax bombs and arson attacks against the owners, the pub in which they drank, their golf club and even their cleaners.

In October 2004, animal rights extremists stole the remains of the 82-year-old mother-in-law of one of the owners from a churchyard in the nearby village of Yoxall.

Her body was only recovered earlier this month after Smith revealed its location in a nearby beauty spot in Cannock Chase to police.

Smith raised a defiant fist to the court room as he was taken down to the cells with his three co-defendants.

Judge Michael Peat told the four that they could have resorted to lawful means to change a law they did not like because they lived in a democratic society.

"You thought to enforce your view not by reasoned debate or lawful protest but by subjecting wholly innocent citizens to a campaign of terror," the judge said.

He said that the Hall family had run a lawful business and said he had read 38 statements taken from victims targeted in the campaign. ...

Well, that scientific stuff pretty much went over my head. I tried to read it, but I just sat here drooling with mouth agape...

Wonder if the doggies can blast stuff with laser beams from the eyes? Are they some kind of super canines? Perhaps they'll fly over Iran and drop little brown bombs on Ahmadinejad's head till he gets so frustrated he'll forget about international jihad...

Does this mean it's maternally inherited? Or something in the nuclear DNA affecting development of mitochondria? Or some combination?

Maybe there's also a genetic explanation why hard core lefties have eyes but don't see, have ears but don't listen and have mouths but don't speak anything true or meaningful...

Perhaps the answer ties in with the old saying about idiocy being empirically defined as doing the same failed thing over and over again and still expecting a different result. :-)

No, Laura - the mutation isn't in the mtDNA, it's causing some sort of disruption in it.

Also - something that isn't getting enough attention by those folks who do "paleogenetics" (tracing lineage through mtDNA to millenia past to declare where sub-species branch off, etc.) - it's recently been discovered that one can inherit mitochondrial dna from one's father.

It's too bad Alien Jeff thought automatically that Kate is necessarily showing off. What I got from the post is that she was talking about one of her professions, mini schnauzer breeding, on her blog, just in case other fellow canine breeders are interested, or geneticists.

Though I'm limited to the basics I learned from high school genetics, I can appreciate the extensive research Kate must've put into learning about genetics as it applies to dog breeding. It reminds me of all the learning and trial and error I've put in (but haven't been into for a while) for my, ah, sometime obsession with bodybuilding. From age 12, I absorbed every bit of scientific info on everything I could from all available sources as I used to plan on following in Arnold's footsteps and all that... don't know what happened to that. I think it was the university thing and the subsequent frustration over the years in not getting my planned career in business jump-started... thanks to a crappy economy and policies like bilingualism and preferential/profiling hiring practices thanks to leftist governments.

Oh, well... sorry for dumping that on y'all, but I just wanted to say how I can appreciate Kate's knowledge of genetic science as it definitely helps with the dog breeding.

Been following the art and breeding for over a year now, Kate. You do excellent work and deserve to take pride in it.

Speaking of work, gotta get back to it... I was just taking a much needed break from a really crazy day in which all sorts of things have gone wrong in my company this week...

Excellent work, Kate. And you don't need a university degree to be scientifically astute and innovative; in fact, in quite a few instances, the university mindset traps you; it locks you into a 'majority mode of thought' which can be quite hostile to innovation and new ideas.

Intelligence has nothing to do with a university degree. I've seen so many PHDs who are the dumbest of the dumb (in my view) - and I have one of those degrees. That includes not only those in the humanities and the social sciences, which are the haven of the irrational and illogical postmodernists, but also, those in the sciences. In science, you'll find so many who fall into a particular perspective and remain locked in it for their entire life.

So- well done, and my congrats. And announcing one's accomplishments is not akin to boasting, alien. But, denigrating someone else's accomplishments IS an act of pure jealousy. So?

I have always believed that people should be complimented and encouraged in whatever good things they may do. Makes for a better social atmosphere generally. Also, sometimes it is better to turn a blind eye to peoples innocent failings.
As far as "HOT" goes, I don't know. I see a picture of an attractive lady on her website, but have never met Ms. McMillan. My wife and I have talked about dropping in at Delisle one day, but always seem to have too much on the go...

Hello Kate,

I am myself surprised to see how a 75% decrease in a major mitochondrial enzyme like NADHD in all cells causes only retinal dysplasia. I would have expected a complex syndrome, including myopathy and encephalopathy. Human diseases caused by a nuclear mutation affecting mitochondria include Leigh syndrome and Barth syndrome. I guess dogs are tougher than men.

I also liked this "test-breeding to define the mode of inheritance". You guys do stuff we are not allowed to do; I'm jealous.

The researchers are still scratching their heads about that - they can't believe the affected dogs are alive.

The other tidbit not mentioned in the abstract - the heterozygous carriers (which are normal eyed) have a mixture of normal and abnormal mtDNA in their cells.
